Clinical Pharmacology Overview

Drug Class: Gabapentinoid

Indications

  • Neuropathic pain
  • Partial seizures (adjunctive therapy)
  • Generalized anxiety disorder (some regions)
  • Fibromyalgia

Mechanism of Action

Pregabalin binds to the α2-δ subunit of voltage-gated calcium channels in the central nervous system, reducing the release of excitatory neurotransmitters such as glutamate, norepinephrine, and substance P.

Dosage & Administration

  • Initial dose: 75 mg twice daily
  • Usual dose range: 150–600 mg/day
  • Dose adjustment required in renal impairment

Adverse Effects

  • Dizziness
  • Somnolence
  • Peripheral edema
  • Weight gain
