Pregabalin
Clinical Pharmacology Overview
Drug Class: Gabapentinoid
Indications
- Neuropathic pain
- Partial seizures (adjunctive therapy)
- Generalized anxiety disorder (some regions)
- Fibromyalgia
Mechanism of Action
Pregabalin binds to the α2-δ subunit of voltage-gated calcium channels in the central nervous system, reducing the release of excitatory neurotransmitters such as glutamate, norepinephrine, and substance P.
Dosage & Administration
- Initial dose: 75 mg twice daily
- Usual dose range: 150–600 mg/day
- Dose adjustment required in renal impairment
Adverse Effects
- Dizziness
- Somnolence
- Peripheral edema
- Weight gain
